Hillcrest Baptist Church began as a mission of the First Baptist Church of Clarksville, Tennessee on March 4, 1964, at the Tarpley Funeral Chapel in New Providence, Tennessee. There were 18 people present at that first meeting. On May 3, 1964, moderator Garner Griffey called the mission congregation to order for the purpose of calling their first pastor John P.S. Humphrey. Pastor Humphrey served until his resignation on August 1, 1971. On September 9, 1964, the Dover Road Chapel Mission voted to construct it's first building. Construction began on September 21, 1964. The name Hillcrest was adopted at the October 28, 1964, business meeting to be used upon occupying the new building.

On October 17, 1965, the Dover Road Chapel was constituted into a Baptist church with 179 charter members. The church doubled the building size in 1966 with a building that included a fellowship hall, kitchen and Sunday School space. On the third Sunday in January of 1972, Marvin E. Hines, Sr. was called as the church's second pastor. Pastor Hines served until his retirement December 31, 1985. The June 1986 business meeting marked the calling of Steve R. Murphree to become the third pastor of Hillcrest Baptist Church. Pastor Murphree resigned in June 1991.
